Richard (played by Pierce Brosnan) is a brilliant poetry professor at Cambridge and an incorrigible womanizer. However, he manages to meet the woman of his life (Salma Hayek) and is convinced to move to California. Consequently, he tries to put an end to his passion for the fair sex and stop his wanderings. However, things are not going as expected. In fact, it turns out that Kate (Jessica Alba), the half-sister of Richard’s college-going sweetheart, has become pregnant.
My Sister’s Boyfriend then follows Richard’s life as it changes dramatically. Upon learning this detail, a series of events occurs and his daily routine is disrupted: marriage, family… and what will become of his history with Olivia (Salma Hayek)?
